Abstract: AIM To prepare temperature dependent hydrogel and to apply it as the scaffold in fabrication of tissue engineering cartilage and to observe the results. METHODS Various concentration and additives of hydrogel were prepared, the gelation temperature were determined, the most favorite hydrogel (250 g*L-1) was selected to be used as the scaffold of tissue engineering, and then the cartilage was fabricated. The chondrongenesis was studied histologically for 12 weeks. RESULTS Gelation temperature of 250 g*L-1 hydrogel was 5℃, and its mechanical properties were good. However the solubility of additives was too low. The 250 g*L-1 hydrogel was applied in tissue engineering; 4 weeks after injection, the hard nubble was formed subcutaneously and immatural cartilage was observed. 8 weeks after injection matural catilage was observed. 12 weeks after injection the amount of matural cartilage increased. CONCLUSION Gelation temperature depends on the concentration of hydgogel, the gelation temperature of 250 g*L-1 hydrogel being 5 ℃. The hydrogel could be applied in fabrication of tissue engineering cartilage, and the cartilage could be fabricated successfully.
