Inhibitory role of genistein in proliferation of human pituitary adenoma cells

Abstract: AIM To study the influence of genistein on the proliferation, DNAsynthesis and cell cycle of cultured human pituitary tumors cells. METHODS MTT method and 3H-TdR incorporation cell cycle analysis were used to examine the changes of proliferation and DNA synthesis of humanpituitary tumors cells. RESULTS In a dose dependent manner, genistein at different concentration (10-7 mol·L-1, 10-6 mol·L-1, 10-5 mol·L-1) could significantly inhibit the proliferation of human pituitary adenoma cells cultured in vitro. The change rate of A490nm value were (91.2±4.9)%, (78.8±5.3)% and (65.8±5.6)% respectively (P<0.01), and the change rates of cpm value were (88.6±3.6)%, (75.5±5.7)% and (58.9±4.9)% respectively (P<0.01). GST (10-5 mol·L-1) increased the proportion of cells in G1 phase from 68.2% up to 91.0% (P<0.01). Moreover, the effect of genistein on the proliferation of cultured human pituitary tumors cells was independent of estrogen receptor. CONCLUSION Genistein inhibits the proliferation, DNA synthesis and cell cycle of cultured human pituitary tumors cells, and its inhibitory effect is independent of estrogen receptor.
