Abstract: 目的: 探讨单一及多位点核酶对乙型肝炎病毒靶RNA的切割作用. 方法:利用计算机辅助设计针对乙型肝炎病毒C基因的三个单一核酶及特异联合型核酶,构建三个核酶各自及三个核酶串联的自剪切转录载体(pGEMRz123), 观察单一及串联核酶(Rz123)对靶RNA的切割作用. 结果: 构建的串联核酶自剪切转录载体体外转录后,在顺式核酶发生自剪切后可将目的核酶正确地释放出来,计算机设计的单一核酶体外可剪切靶RNA;串联核酶中的单一核酶不仅可单独作用,而且可联合切割,提高了单一核酶的剪切效率. 结论: 抗乙型肝炎病毒特异联合型核酶体外可联合切割靶RNA.
